I am not quite sure what sort of "weight" you are refering to. If you mean 
"baggage" I can recommend to place it as low as possible (e.g. at the wheel 
hub). It doen't make cyling easier but it makes it safer ! The lower center 
of mass gives a better driving stability. The aerodynamic effects are 
difficult to predict but I guess it doesn't make much of a difference. 

Maybe, you are talking about bike design. A mass at the wheel rim travels a 
longer path that a mass at the wheel hub. Thus, its average velocity is 
bigger and so is its kinetic energy at a given velocity of the bike. This 
means that you need more work to accelereate the bike. (see "rotational 
moment of inertia" or "ratational energy")
