Hi Rob,
There are many many published circuits for what you need to do this 
project. You are asking to build what is just one element of a lie detector 
system, something that measures skin resistance. The more you stress, the 
more you sweat, and the lower your skin resistance becomes. Electrical 
resistance measurement can be done in many ways, applying a fixed voltage 
and carefully measuring the voltage across a sensing resistor is eminently 
practical. You can find circuit examples in many of the circuit ideas 
collections of magazines like Elektor and Practical Electronics.
