Met-enkephalin is synthesized enzymatically in a sequential manner. 
Carboxypeptidase Y is used in the stepwise synthesis of met-enkephalin.

Ac-Tyr-OEt => Ac-Tyr-Gly-NH2 with Carboxypeptidase Y, Gly-NH2, pH 9.0

Ac-Tyr-Gly-NH2 undergoes deamidation to produce Ac-Tyr-Gly-OH

Ac-Tyr-Gly-OH => Ac-Tyr-Gly-OEt with ethanol/HCl

The process is then repeated with Carboxypeptidase Y, (an amino acid)-NH2, 
pH 9.0 until the necessary amino acids have been added, to produce met-
enkephalin:

Ac-Tyr-Gly-OEt => Ac-Tyr-Gly-Gly-Phe-Met-OH with Carboxypeptidase Y, (AA-
NH2), pH 9.0
