Hello Hansrainer: It is my experience that when one does a transplantation all organs of interest are taken at that time. Since death occurs when the integrity of the system is disrupted it would only make sense to do all experiments when the initial sacrifice is done. For instance when transplanting bone marrow cells from one mouse to another, the cells are isolated from one mouse which is sacrificed before the removal of bone marrow. The cells are prepared for transplant and the transplant must take place in a timely fashion. I cannot say for certain in your Spemanns Transplantation Experiments, but I can only go on what I have experienced. When we want multiple samples from one animal, we take them at once and freeze upon removal in liquid nitrogen. This is for metabolic analysis. In the case of transplantation the logistics would be complicated with more than one tissue. I suspect that the remainder of the embryo is discarded.
