Subject: risks in drugs tested on animals given to humans at 100 times the dosage

Why would it be a risky thing to give a larger dose of a drug to a human, 
that was tested on a guinea pig.  I realize the metabolism is different 
but not sure why it would make a difference.  Any answers would be greatly 
appreciated.
