Dear Stuart, I know this project well, I did it myself in the final year of High School last century. To change the pH you might need. acid (0.1M hydrochloric acid) alkali (0.1M sodium hydroxide) The buffers test tubes (quite a few large ones) measuring cylinders (two, one for acid one for alkali) electronic pH meter Ok, firstly you set up the solutions: in test tube one put in about 10 ml of acid, and 2 ml of pH 2 buffer (check using the pH meter, if different add acid/alkali to get to the level) then in the rest 2: 9ml acid, 1ml alkali, 2ml pH3 buffer 3: 8ml acid . 2ml alkali, 2ml pH4 buffer 4: 7ml acid, 3ml alakali, 2ml pH5 buffer, and so on...... that should give you nine test tubes with different pHs in them for you to conduct the experiments. Good luck with the project and thanks for asking. And have you tried changing the concentration of enzyme ? yours scientifically, Peter D. Traill
